Evaluation and results
The Aklides Kyto system was the first immunological product for primary screening and the analysis of cell-based immunofluorescent assays; it can analyse Hep-2 cells as well as granulocytes (ANCA), crithidia and tissue sections. Automatic image acquisition of the preparations on the object slide is done by an 40X lens. The preparations are focalized in the blue DAPI channel to which a green FITC channel is added for contrasting staining.
This contrasting staining facilitates robust focussing of positive as well as negative preparations. Every preparation is typically examined at 5 positions and documented by DAPI/FITC multichannel pictures. Analysis of the preparation takes place during measurement on the basis of the image data. At the same time, qualitatively insufficient areas, for example due to staining artefacts, are excluded from analysis by algorithms.
Cellular structures (e.g. the nucleus) are detected in suitable image regions and morphologically analyzed by efficient image analysis algorithms.
Subsequently the FITC channel signal is determined in morphologically adequate image regions and structures.
